Hi,

I'm purifying HIS-SUMO-GST protein, the expression is done under 37C for 4.5 hr and the protein is purified using Nickel column followed by gel filtration. I want to keep the GST tag with the SUMO, but then there is self cleavage of GST.

any ideas for preventing the GST from falling off itself?
