I have a couple of suspension cell lines that I am thinking about using for developing a binding assay--I wanted to run it sort of like an ELISA with my cells stuck to the plate and after adding my protein I will wash and add conjugated reporter antibody. The problem I am having is I can't find any adherent cell lines that work--I am focusing on HER2 or CD20 lines and they would all be bone marrow derived cells. Is there any way I can adapt a cell line to grow adherently? If not, is there a coating I can put on my plate that will induce them to bind to the plate strong enough to allow me to wash unbound protein from my plate before I add my reporter antibody?
