Hi,

I am currently working with THP-1 cells. I differentiate them with PMA overnight, wash off the PMA and then infect the adherent macrophages. Typically  my experiments last between 2-3 days. I am now starting to look at introducing dominant negative plasmids and siRNA into these cells. During  literature search, I  have come across protocols that involve  transfection of  these cells both before and after differentiation. I am not sure which protocol is the best to use. I would appreciate any advice I can get on how to transfect these cells!!
