I am using E coli DNA for my experiments. What I need is to restrict 24Kb E coli DNA to two equal halves and then to label one half (24Kb) DNA with biotin and DIG. Then I attach this DNA with streptavidin and anti-digoxigenin coated beads. My problem is that the protocol that I am following doesnot give me strong enough coupling between the beads and DNA and the binding breaks very quickly. Can anyone suggest where to find detailed protocol. I would appreciate any help. Help on preperation of beads will be also very much appreciated.
