I am trying to look at aconitase activity in mouse brain and heart. I am using a kit to isolate mitochondria and then lysing mitochondria with CelLytic M. Mix lysate with substrate buffer and incubate at 37 in microplate reader for 15 min. Then read every minute at 340nm for 15 min. I am getting some absorbance but it remains at the same level throughout the 15 min.

If anyone has every done this assay, I would love to pick your brain.
