Does anyone have photos of primary mouse myoblast culture at various stages? Are they supposed to be fibroblast like or do they remain as little spheres for a while (while the fibroblasts in the culture quickly adhere and spread)? Does it make a difference what you plate them on as some different protocols suggest gelatin, collagen, ECL and similar matrices? We're going to try differentiating some more as our first attempt was unsuccessful. I don't have the desmin antibody normally used to identify them by immunofluorescence, but would antibodies for Pax7, MyoD, Myf5 or myogenin work? Any others?
