Hi!
While transfection of cells with a plasmid containing the GFP gene, I observed cells under a fluorescent microscope, bright fluorescence as well as the damped. Can somebody help me, hence the difference?

Do you mean that you saw fluorescence in the untransfected as well as the transfected?  If so, you probably have either autofluorescence or didn't wash out the fixative enough.
