Hi,
i'm trying to express a protein which have high pI value (predicted to be 9.8) in e.coli expression system. after lots of optimization (inducer, temperature, different fusion tags and e.coli strain). the protein cant be expressed. the sequence was ok (100% same as the original sequence). is high pI can effect in the expression..should i consider other expression system? thnks a lot!!
anyone have experience expression high pI protein in e.coli
