I´m trying to clone some genes in a plant vector, at the time that I use the CIAP enzyme in order not to obtain religation and the T4polimerase to fill up the genes with dNTPs so I can put the genes into the vector. When I transform my ligation reaction using some E.coli colonies, at the next day I don´t have any colony grow. What I´m doing wrong?
