Hi everybody,
I am new here and I need your help. I am thinking to do a project concerning antigen presentation of dendritic cell.  But I do not know how I should load DC with peptide. Is it enough to add peptide in medium? should ı use serum or not? What kind of things  should I consider esp .peptide design and medium ingredients? Please help me. If you have any protocol for loading  DC with antigen  and share here, İt will be so good ...

Usually it is enough to add peptide to medium. But one should optimize loading time and %serum (you can start with overnight/ 5%FBS). First you can try loading with GFP and check it by flow cytometry.
