I am doing superoxide anion measurement. As i refer some articles, it's using Krebs - HEPES solution to keep the tissue before run the assay, some is oxygenate the tissue, some directly put the tissue into cold Krebs-HEPES solution. And i had been told thet the Krebs-HEPES is used in the determination of intracellular and extracellular released of ROS and NO. I would like to ask that why it is specially used in the determination of intracellular and extracellular released of ROS and NO? And, i also face some problem in making this solution, the solution tend to become cloudy (not clear) when i put in the CaCl2. 2H2O, is it any solution for this problem? Thank you.
