Hi All,

I have cloned some high GC content DNA (64% approx) from Leishmania parasite in pUC18 and transformed in XL1-Blue. The insert is getting lost during next round broth culture or plating. Can any body suggest some remedy?

I also clone GC rich DNA (70%) and met the same problem, may be the unclonable  DNA have secondary structure.I want to try another host strains such as SURE (stratagene) or STBL2 (GIbico)
