We normally use the qiagen puregene kit for our mouse genomic DNA extraction (tail clips). I have 2 mouse hearts (one from a null, one wild type) in a jar of 10% formalin. I know, don't ask... Anyway, can I take a small piece of heart and use the puregene kit to digest and purify DNA to re-genotype? If not, are there other protocols for doing so? Should I immediately take them out of formalin (we normally store them at 4 deg C that way indefinitely) and put in PBS to stop anymore crosslinking?
