Hello, I've been stuck on this for some time, and can't find a definitive answer. I am culturing mouse splenocytes to assay antibody production after stimulation with LPS alone or in combination with cytokines. I understand that LPS+cytokines induces isotype switching to specific isotypes, but what does LPS do alone? I've read many times that it both causes polyclonal B cell proliferation (in high doses) and induces class switching, but does it cause switching to specific isotypes? The data I currently have is not very reliable, and papers I've seen only consider LPS in combination with other factors. Do any of you have any experience/knowledge in this area? I would appreciate any help/advice you could provide, thank you!
