Help to construct luciferase DNA construct

Hi
   we have HCV genes expression vectors and we have screened siRNA activity against it. Now I want to make their luciferase DNA constructs to screen against shRNAs. Can any body tell me how should I proceed|?

Since you are doing RNAi using siRNA or shRNA, you can simply measure your target gene expression and don't need to put the target into a reporter (3UTR).

Thanks pcrman for your kind reply. You are right but when we see literature regarding shRNA silencing, in almost most of the literature people have mostly measured the inhibitory effect of shRNA by making DNA constructs in luciferase reporters while I have the cloned the structural genes of HCV in PCR 3.1. Can you kindly suggest how i should proceed?
