i cloned viral P protein into the T easy vector which gave successful colonies on lb ampicillin agar. i then isolated plasmid and found it of the weight higher than that of the vector i then did a restriction digestion of the vector which released the fragment of appropriate bp. i wanted to do pcr jus to confirm . but it faied time and again. there is a 200 bp band that means the PCR is working then why is the fragment not coming?
