why? cos you got only a weak signal after O/N incubation? or you've to leave the blots over the weekend? or you forgot the blots in the Ab solution over the weekend? ;) there's only one way to find out....so give it a try but you might get higher background.

I think so, it should be fine if you leave your blots with primary antibody in cold room for two nights instead of one. I am telling this because I have done this for my primary antibody and it didn't create any problems while developing the blots.

If it's supposed to bind it'll do within 4 hours. 2 nights is not necessary
