Spiking produce

Hi there,
I have an experimental outline that states sampling from Day 0, 3, 6, 9 etc. On day 0, am I plating the samples before I spike them?  Or does Day 0 take into consideration the spiking?

The way I would understand that would be to spike, and then the day 0 would be plated immediately after spiking.
