Hi,
I am trying to optimize conjugation with HRP or Biotin for the mouse monoclonal antibody. I use Pierce protocol for biotinylation. For HRP, I used periodate to activate HRP and conjugate with the antibody. But the problem now is the antibody titre is low in Elisa. I am trying to improve its sensitivity. Anybody can give hints to improve the binding capability?

All components of the conjugation procedure have to be optimized.
To increase binding a quick test would be to titer PEG in your immunoassay. It will increase binding...you may also have to increase number of washes and/or amount of surfactant in the washes.
