I am encountering a problem in SDSPAGE
Details
when we run the gel without boiling the sample in SDS gel loading buffer we do not get any protein bands in the lane except at the bottom of the gel, where a thick band is observed.whereas if we boil and load the sample as per the std. protocol for 2 min. we get a kind of protein ladder with diminishing of the low mol. wt. chunk.
what could be the reason
