BM Purple

We have been using the AP substrate Bm Purple from Roche for detecting mRNA probes in tissue sections. Occasionally, the color reaction goes awry!!! The normal dark blue/purple color is instead turquoise....Beautiful but confusing and not so useful.

Our sections of paraffin, 6 microns thick, from human tissue (lung and gut). We use digoxigenin labelled probes, follow standard, published in situ protocol.

Roche has no suggestions. Do you????????
