Verifying insertion of homologoous recombination

Let's say I want to homologously recombine a gene of interest into a particular gene locus in eukaryotic cells. I design my vector to have a 5' homologous arm followed by my gene of interest, a positive selection gene (eg G418), and a 3' homologous arm, and downstream of the 3' homologous arm I have a negative selection gene(tk). What is the easiest method to determine that my vector only homologously recombines where I want it to?
