we want to extract DNA from smears: some of smears are old and coverd with cover slides ,we put slides in xylen to seperate cover slide sometimes for 3 or 4 dayes,Is xylen damage DNA?
we use Nucleospin kit and QIAamp kit for DNA extraction ,we can not get good concentartion of DNA.
when we measured DNA concentration with Nano Drop we see ratio 280/260 around 3 or 4 ?what is mean?and strang curve.
Does anyone any suggestion ?
what should I do?

u mean u want to extract DNA from smears on slides.
ok, well:
xylene should not damage ur DNA ...
since when going for extracting DNA from paraffin embeded tissues, we add xylene ..
so, no worries with xylene ...
u mean 260/280 ?
A 280 : protiens with aromatic compunds
A 260 : DNA.

yes,we extracted DNA from Bone marrow smears ?but we dont get good result yet
i attached some of curves of nano drop result
we did WGA and clean up then repate with nano drop but so bad result again

xylene should not damage ur DNA ...
since when going for extracting DNA from paraffin embeded tissues, we add xylene ..
