Use of Nonidet to treat faecal samples

Hi,

Wondering if anyone has any experience in the use of Nonidet P40 for the treatment of faecal material? I want to treat the faeces with Nonidet to remove the envelope from any virus particles in there, and to lyse any cells to release intracellular virus. I've found some rather old papers (inc. attached 'STRUCTURAL POLYPEPTIDES OF THE MURINE CORONAVIRUS JHM' by Wege et al.) which describe using 1% (v/v) nonidet P40 and incubating the sample for 20 degrees C for 15 minutes. Does anyone have any advice on:

How much Nonidet to use - I make a 10% faecal suspension in HBSS, so do I add Nonidet to 1% of the total suspension or 1% of the amount of HBSS I added?

The Nonidet comes as a 5% aqueous solution - presumably when papers refer to using 1% nonidet they mean 1% of the 5% solution as Nonidet P40 seems to be a trade name?

Can't find anything more recent about it's use, any help much appreciated :-)
