Dear Colleagues,
I have been transfecting expression plasmids into mammalian cells now for about 3 years and suddenly now I have this question that I cannot find an answer for....
1. How does the vector sequence encoded in the plasmid transcribe in the mammalian cell (after transfection)? as far as I know RNA polymerase II is only in the nucleus of eukaryotes, then how does this sequence get transcribed to become mRNA?
2. When we say 'stable transfection' it is meant that the plasmid has now integrated into the host genome. How does this happen? What sequence information does the plasmid carry for the vector sequence to be integrated into the genome of the host cell?
Any clues to these questions will be of great help.....I just can't get sleep till I find an answer for these questions....:(

thnx
