I have established some stable TetOFF cell lines using Hygromycin selection, and would like to confirm integration of this cassette into DNA by southern hybridization. I have tried a simple Hygromycin ORF probe on Phoenix cells (which should be Hygromycin positive), but no signal. Does anyone know a probe (or a website where I can design one) against Hyg or tTA?
