Hi all
I have to use pH sensitive fluorescent probe for determining the intracellular pH of my cells in experimental conditions. I decided to use SNARF-4 AM ester. The procedure seems relatively simple using a fluorescence plate reader. However, I am having a hard time finding a good protocol for doing the calibration. Literature has several different method and I have chosen Nigericin calibration with high K+ buffers. However I can't seem to find the recipe for making the buffer. Anyone has any experience using those or can help me figure out? Help is appreciated.
