I would like to know some methods to measure the exact concentration of my protein ( Nisin). Can I find some help from you.

purified or as part of a mixture?
if purified then you can perform one or two of any number of protein assays, depending on concentration.
one of the more exact assays is based on the kjeldahl (or micro-kjeldahl) nitrogen determination.
if part of a mixture then you can determine the concentration of the mixture then determine the percentage of the mixture that is your protein (by page or gel filtration).
