I'm trying to induce the myogenic differentiation of NIH3T3 cells. This is the paper I used as a reference: http://www.ncbi.nlm....ed?term=2748593 . However, there seems to be something wrong with my differentiation media as all the cells died on the second day after I fed them with differentiation media. Currently, I am using DMEM + 5ug/ml transferrin + 10 ug/ml insulin as my differentiation media. The DMEM I used is high glucose (Invitrogen, Contains 4,500 mg/L D-glucose, and L-glutamine, but no sodium pyruvate). The insulin is "insulin, human recombinant, zinc solution" from invitrogen whereas the transferrin is "Transferrin Human" (T8158) from Sigma.
