Dear all,

I have a few questions on sequencing.

When reading on the new sequencing techniques they often speak of 3the generetion sequencing and they call it single molecule sequencing, but what does this mean?
Do they mean that with this third generation (like helicos) you do no longer need to PCR your DNA? That you can read the DNA sequence directly from the template DNA without the need to amplify your samples? Or is there another explenation?

A second question I do not get is why the quality of the first bases is bad (and also the quality of the last bases).
I do not understand why this is the case.
