here's my situation: I have to clone a 250bp fragment in a 9400 bp length vector. though one ha?
I was wondering if any one can help me with it. any suggestion or hint is deeply appreciated.

What is tough about it? I would suggest digest - purify - ligate. But maybe I am missing a part here... Could you deliver a bit more details?
