Would anyone have a protocol for staining RBC in vivo with CFSE? Is this possible, or should I give up and instead stain them ex vivo? The issue is that I require ALL RBC in the model to be labelled with CFSE.
My attempts at in vivo staining of RBC have resulted in very low levels of labeling, barely above background (with is with 10uM). I have a feeling that I may need to increase the concentration of CFSE, but worry about toxicity. The lymphocytes on the other hand, stain very brightly in vivo.
