HI ALL

I want to know what is this normalization of the data in densitometric analysis for the semi-quantitative expression analysis.I am using this IMAGEJ software but i am little confused as how to represent the data. Why do we have to plot the graph against the control? Can anyone plz explain the exact procedure step by step to do this.

THanks in advance

You first obtain the densitometric numbers for your gene of interest and also housekeeping gene, then normalize GOI density to that of the HKG for each treatment or sample, then plot the data as fold changes to a control treatment in a bar figure.
