I'm studying a virus which induces apoptosis in multiple myeloma cells (as measured by loss of viability as well as cleavage of Casp3 and PARP). I tried to inhibit this apoptosis with a caspase 3 inhibitor Z-DEVD-FMK at 100uM (the highest concentration recommended by the company), preincubation of cells with drug for 1 hour followed by virus infection, and analysis of apoptosis using CASP3 and PARP cleavage at 24 hours. I saw virtually no reduction in cells death and only a very small reduction in cleavage of PARP using the drug.
Anyone ever use this drug before? Does the drug just not inhibit caspase-3 100% or is there non-caspase-3 mediated cleavage of PARP?
