cell synchronization

i would like to study the effects of a gene on cell cycle. since my cell line is difficult to transfect, i used lentivirus to infect the cell line and did stable selection. i synchronized the cells with nocodzole, and i found the percentage of cells arrest at G2 phase by nocodazole between the cells infected with vector control and my gene was different, even though i had tried several time points.

i would like to know what i should do to solve this problem. is this approach correct?
people do not claim they have done synchronization in papers, and is it necessary to do synchronization??
