I had tried several times to extract DNA from Mycobacterium by using either commercial kit and phenol chloroform extraction method.
However the DNA yield and concentration is not satisfied.
Exp: 1ml of bacteria culture, i could only get 0.5ng, A260/A280 ratio around 1.4. (i need 10ng of DNA)
Did anybody experience this? how can i improve the yield and purity?
