I used lentivectors(pCDH-MCS-T2A-copGFP-MSCV (SBI)) for cloning CD40L, 4-1BBL & chimeric immune receptor anti her2.
Vector sizes is about 7.1kb & Insert sizes are about 750bp, 780bp & 1550bp, respectively.
I inolucted 2ml of LB liquid media & incubated for 18hrs (at shaking incubator)
& did miniprep, but average yield was about 40-50ng/ul.
Do you think what is the main problem?
I want your precious opinion about this problems, which will be very helpful to me.
P.S: The strategy to add Chloramphenicol before harvesting will be helpful?
