has anybody done SOD activity assay by the method of Kono (1978)??
it is calculated as amount of enzyme inhibiting NBT reduction by 50%. i dont understand the funda of 50% inhibition??

Download the information booklet for the Cayman SOD assay kit for a better explanation than I can give. They use a different superoxide generator but the principals of detection and quantification are the same.
I’ve found that I need to run an antioxidant control (epicatechin is good) as well as a SOD standard in order to correct for inter-assay variability.
