I'm wondering if anyone here has done nested DNA deletion cloning using Exonuclease III followed by Mung bead or S1 nuclease? This looks like a very intriguing technique, but I'm wondering if the Exo-3 digestion can really be well controlled such that it won't overdigest?
I guess I'm looking for if someone has had good or bad experiences with this technique it would be great to know (e.g. "I've tried it before, doesn't work well at all, don't waste your time" or "It's a great technique, very consistent", etc).
