I have always used methanol at -20C for HEp2 cell fixation when grown on slides, but it doesn't give as good activity as acetone. Acetone does not retain nuclear morphology for mitotic cells. Does anyone have a fixative or process that will allow me to maintain mitotic cell morphology and high reactivity for IFA/ANA testing.
