I am trying to do MTT assay using HL 60 cells .The idea is to testing the activity of natural product on the HL 60.I have been trying to standardise the protocol but no chance,which is:

plate 20,000 cells/well
add extract ,incubate for 3 days
Add  50 ul MTT ,incubate 4 hrs
Centrifuge 3500 g  for 10 mins
Discard medium ,add 50 ul DMSO.
Shake 5 mins ,read

Can anyone help  me( or improve ) with this protocol
