hi admin and chatter,

i would like to asking about PCR-RFLP scoring, i'm currently doing genetic variation of fish species, i;m using several RE,

i have pattern of RFLP as below:

Sample AciI Cvk Hpy Composite genotype
1 A A B AAB
2 B B A BBA
3 … … … ….

i;m little bit lost. based restriction patterns we produce by given enzyme identified with latters A,B,C and so on according to the individual pattern, were then interpreted as the presence or absence data. that what i know. but my question is by given the restriction enzyme identifies with latters A, B and so on, should we know the size fragment of each pattern of RE, or just give pattern based on what band produce and ignore the size fragment of RE. i;m thinking if we dont know the size fragments the band produce could be a "ghost band/ star activity"

i hope u can give your opinion and some advise.

thank you
