I am trying to amino acid starve some human cancer cell lines.  I heard EBSS (Earls balanced salt solution is good to use.   My question is, should I add 10% FBS to the EBSS or just use the EBSS by itself?

FBS contains all sorts of stuff, including proteins which the cells can break down into component AAs...

If you think you really need to use FBS, consider dialyzing the serum against PBS. We do that in cells that we label for pulsed SILAC
