Hello,

I found this protocol on google. It basically skip the gel extraction step, and directly use the DNA that was still in agarose.

Anyone has tried this please let me know. I am in the process of finding a good strategy for my cloning, I havent started anything yet. Is this shortcut work ?

I know people who do it, and it seems to work OK.

so using this shorcut, you would ignore the calculation part ... no more molar ratio btw plasmid and insert? I d like it if it does work

I guess, though you could calculate the molar ratio given the amount of input...
