I have a question concerning cloning the lysyl oxidase gene (LOX). The pre-LOX protein is glycosylated in the Golgi and is a 50kDa protein. It is then secreted into the extracellular matrix where it is cleaved into a 30 kDa active enzyme and a 18kDa propeptide at a Gly/Asp bond. I need to isolate both genes for the 30kDa active enzyme and 18kDa propeptide. How would I achive this? If I clone the whole LOX gene, will it be processed into the 30kDa and 18kDa proteins? If so, how do I isolate the two genes?
