Suppose we digest a plasmid with asc1,then after seeing the gel pictures we got a single band with some samples..My question is simple in what condition we normally go for double digestion. my supervisor told me to digest with hindIII and sac1 after seeing the gel photos of asc1 digested sample..I asked hm the reason bt hs explanation was confusing...please i will appreciate the help..if u require the photos to see i will send u later
