Having problems in getting gigaseal in patch clamping

Dear All


I have recently started working on cultured hippocampla neurons. I will be doing inside out single channnel patch clamp recording. Its been a week in the lab but I have not been able to get a giga seal yet. On a couple of occasions, I got it but the membarane broke soon.

I need advice on my techinique like teh angle to approach neuron and the area of neuron to targI am sure the rest of the set up is fine as ppl in the lab have been able to so in the recent past. We are sylgarding, fire plolsihing, resistance is ok, solutions are filtered etc.

Any help for newbie in patch clamp woul dbe appreciated. I have previously worked on two electrode volatge clamp in xenopus oocytes.
