Primary hippocampal neurons seem to be quite insensitive to glutamate treatment. Does anybody have a protocol treating them in magnesium-free, glycine containing medium to increases their sensitivity (disinhibtion of NMDA receptors)?

The time of the experiment can influence the toxicity. Hippocampal neuron are quite insensitive to glutamte within the first 5 days in culure.
This experiment is usually done in Locke's solution and you have to add glycine separately. The quality of these solutions can make a difference.
