Tried to extract RNA from 5E5 human fetal spleen and kidney cells. Been using the qiagen RNeasy mini kit and trizol. The quantity was so small, <0.06ug/ul. A260/280 was about 1.5-1.6. The cells were stored in liquid nitrogen for 4-6 years. THe cells were thawed and washed with cold saline twice in eppendoff, then either RLT or trizol were added. How can I improve the yield and purity. Thanks for help.
