293 T cells

Can anybody let me know whether 293T cells are attached (adherent) cells. Is there any need to coat the flask with Poly lysine for these cells. How many passage can be done with those.

Yes they are attached and you shouldn't need to coat the flask. They do not attach very firmly, so can often be washed off with pipetting if you are not careful.

Dear Sanku,

Bob1 is of course correct, HEK 293T cells should be adherent. However I have grown these cells alot and have noticed that the adherence is variable depending upon the TC plastic you use. We generally used Becton Dickinson (BD) which gives us the best adherence. This is very important as just washing the cells with PBS prior to trypsinisation CAN also wash off the monolayer of cells

BUT this can be very useful for example if you want to do membrane receptor studies and NOT use trypsin to release the cells from the plastic.
